Quick explain levadopa:
Prodrug that is used because it can cross BBB, there it can be converted into dopamine to treat Parkinson’s
Big problem is that most goes elsewhere outside of CNS and cuases side effects
What drug blocks AAAD(Aromatic Amino Acid Decarboxylase)?
Why is this advantageous for Levadopa therapy?
Carbidopa!
Inhibits AAAD in periphery and does not cross BBB, this is great becuz then it doesn’t interfere with action in the CNS
What is the carbidopa/levadopa combo called?
Sinemet!
What are the adverse effects of Sinemet therapy?
Dyskinesias - involuntary movements occur
On-Off Phenomen - Rapid random loss of drug effects
Wearing off - over time occurs between doses

What is entacapone?
it is a COMT inhibitor
This drug is meant to decrease the peripheral metabolism of L-DOPa by COM-T.
What is the drug, Selegiline used for?
It inhibits MAO-B
This keeps dopamine in the sympase rather than having it eaten up by glial cells
What is pramipexole used for?
It is a D2 receptor agonist
So it bypasses the whole L-Dopa conversion thing
What kind of drug is Trihexyphenidyl?
It is a muscarinic antagonist
What is a drug to treat parkinson’s that is actaully an antiviral?
Amantadine
