Metabolized by P450 system. Many drug-drug interactions
SE: renal dysfunction, hypertension, neutotoxicity
Like cyclosporine but inhibits calcineurin by binding to FK506 immunophilin
Tacrolimus
Its binding to cyuclophilil inhibits the IL-2 signalling pathway; toxicity effects include hypertriglyceridemia, hepatotoxicity, diarrhea, and myelosuppression
Sirolimus
Mycophenolate mofetil
Purine antagonist
Blocks de novo GTP synthesis by inhibiting inosine monophosphate dehydrogenase
Organ transplantation, GVHD, some autoimmune disease
Oral, parenteral
SE: GI disturbacnes, myelosuppression
Thalidomide
Complex immune effects including reduction in TNF-a production
USed in erythema nodusum leprosum, multiple myeloma

SE: teratogen, somnolence, peripheral neuropathy, neutropenia
